HNPP is an autosomal dominant inherited neuropathy caused by a deletion of one copy of the PMP22 gene on chromosome 17p. It is the reciprocal genetic change to the CMT1A duplication. HNPP causes recurrent episodes of numbness, weakness, and muscle atrophy triggered by minor pressure or trauma to peripheral nerves.

Who Should Consider HNPP PMP22 Deletion Testing?
Individuals with recurrent, painless episodes of focal weakness or sensory loss affecting individual nerves, particularly at common compression sites such as the elbow (ulnar nerve), wrist (median nerve), or fibular head (peroneal nerve). Those with nerve conduction studies showing conduction block or focal slowing at entrapment sites with background mild generalised slowing.
HNPP follows autosomal dominant inheritance. Each child of an affected individual has a 50% chance of inheriting the deletion. Genetic counselling is recommended to discuss the implications for the individual and family members.
